Conserved Region 3 of Human Papillomavirus 16 E7 Contributes to Deregulation of the Retinoblastoma Tumor Suppressor

Abstract: The human papillomavirus (HPV) E7 oncoprotein binds cellular factors, preventing or retargeting their function and thereby making the infected cell conducive for viral replication. A key target of E7 is the product of the retinoblastoma susceptibility locus (pRb). This interaction results in the release of E2F transcription factors and drives the host cell into the S phase of the cell cycle. E7 binds pRb via a high-affinity binding site in conserved region 2 (CR2) and also targets a portion of cellular pRb for… Show more

“…The surface-exposed mutants within CR3 of HPV16 E7 were previously generated by site-directed mutagenesis and are described elsewhere (32). GFP-fused E7 constructs for HPV16, HPV6, HPV11, and HPV18 were described previously (33). Full-length E7 genes from HPV31, HPV33, HPV39, HPV45, HPV52, HPV55, HPV58, HPV59, HPV67, and HPV74 were PCR amplified from the viral genomes and cloned into the pCANmycEGFP vector by using EcoRI and XbaI restriction sites.…”

“…Samples were subjected to coimmunoprecipitation as indicated. patch on the surface of E7 CR3 (32,33). The identification of E7 mutants unable to bind p190 provided us with reagents to study the significance of the E7-p190 interaction, as described below.…”

“…Next, the regions of E7 important for enhancing IL-18BP production were identified. Primary keratinocytes were transduced with retroviruses expressing a panel of E7 proteins containing mutations in the amino-terminal disordered conserved region 1 (CR1) and CR2 or in the more structured carboxyl-terminal CR3 (25)(26)(27)(28)(29). Figure 2A shows a schematic diagram of E7 and the mutations examined in this study.…”

“…This region contributes to a number of E7 functions, including dimerization (36). It may also mediate interactions with a number of cellular proteins, including cullin 2, HDAC, pCAF, p300, TATA binding protein, and Rb (25,27,29,37,38). The interaction of E7 with one or several of these proteins may be important for regulating IL-18BP production.…”
