Conserved Water-mediated Hydrogen Bond Network between TM-I, -II, -VI, and -VII in 7TM Receptor Activation

Abstract: Five highly conserved polar residues connected by a number of structural water molecules together with two rotamer microswitches, TrpVI:13 and TyrVII:20, constitute an extended hydrogen bond network between the intracellular segments of TM-I, -II, -VI, and -VII of 7TM receptors. Molecular dynamics simulations showed that, although the fewer water molecules in rhodopsin were relatively movable, the hydrogen bond network of the ␤2-adrenergic receptor was fully loaded with water molecules that were surprisingly i… Show more

“…This hypothesis was recently validated by the high-resolution antagonist-bound A 2A AR crystal structure that identified the highly conserved aspartate residue in TM2, Asp 2.50 as a key residue within the sodium-binding pocket (Liu et al, 2012). Furthermore, TM2 is involved in a water-mediated hydrogen bond network with TM1, TM6, and TM7, which has been suggested to regulate GPCR activation (Rosenkilde et al, 2010;Nygaard et al, 2010). Our data suggest that the altered conformation, stabilized in the presence of rigid (N)-methanocarba 59-Nmethyluronamide nucleoside derivatives with elongated C2 substituents, may contribute to cytoprotective signaling in A 3 -FlpIn-CHO cells.…”

“…However, the role played by this highly conserved N 1.50 in the activation mechanism of the AT 1 receptor appears to be different from its role in the activation mechanism of the ␤ 2 -adrenergic receptor. Indeed, the loss of the polar side chain decreased the constitutive and the agonist-induced activity of the AT 1 receptor, whereas it did the opposite to the ␤ 2 -adrenergic receptor (24). Furthermore, the MD simulations suggested direct side chainside chain interactions between residues D74 2.50 -N295 7.46 and D74 2.50 -N46 1.50 in the AT 1 receptor, whereas the corresponding interactions are water-mediated in the ␤ 2 -adrenergic receptor (24).…”

Thus, depending on the receptor, the conserved polar residues forming the H-bond networks can act in different ways and through different means.…”

“…Either of the water molecules in the refined models deviate more than 0.75 Å from their original positions, and rotamer states of side chains involved in the water-mediated hydrogen bonds remain unchanged. In addition, GluII:10 (2.50) interacts with Tyr-VII:20 (7.53) through hydrogen bond interactions with a single water molecule, in contrast to the interaction of AspII:10 (2.50) with TyrVII:20 (7.53) in the inactive structures of A 2A AR and B2AR, which is mediated through two water molecules (5,10,51).…”
