Considerations for Size, Surface Charge, Polymer Degradation, Co‐Delivery, and Manufacturability in the Development of Polymeric Particle Vaccines for Infectious Diseases

Abstract: Vaccines have advanced human health for centuries. To improve upon the efficacy of subunit vaccines they have been formulated into nano/microparticles for infectious diseases. Much progress in the field of polymeric particles for vaccine formulation has been made since the push for a tetanus vaccine in the 1990s. Modulation of particle properties such as size, surface charge, degradation rate, and the co‐delivery of antigen and adjuvant has been used. This review focuses on advances in the understanding of how… Show more

“…Finally, translational materials must be easy and inexpensive to manufacture at scale to enable urgent responses to global demand 243 . At every stage of the design process, it is important to consider whether controlled delivery technology could be simplified without sacrificing efficacy and safety 218 , 244 .…”

“…When comparing NPs, there are several physiochemical properties that can vary from platform to platform, but also influence how the NPs interact with APCs and other immune cells ( 22 ). Of the physiochemical characterization of NPs for vaccines, surface charge, particle size, loading capacity of the particles, and drug release are the most commonly reported.…”

“…An important control in the evaluation of innate activity is a vehicle-only control since high production of cytokines can indicate endotoxin contamination. Endotoxin contamination cannot be removed with filtering, but can be easily determined by absorbance with the limulus amebocyte lysate (LAL) test that can quantitatively measure the amount of endotoxin in a sample ( 22 ). Uptake studies by APCs are also an important in vitro assay that is best performed using fluorescent dyes (e.g., fluorescence activated cell sorting/scanning (FACS)) and confocal microscopy ( 25 , 26 ).…”

“…Nanoplatform infrastructures provide improved efficacy in relation to conventional vaccines and marketable tools to fight infectious diseases involving negligible financial risk. Although not the panacea, the future of vaccine research and development may pass through nanoplatform-based approaches for many pathogens [ 75 , 93 , 94 ]. NPs have different roles in vaccinology mainly as adjuvants, carriers, and platforms.…”
