ABSTRACT:In the course of our investigation on biologically active substances from inedible mushrooms in Japan, Germany, and Vietnam, we studied the chemical constituents of 22 species belonging to five families: Scutigeraceae, Polyporaceae, Xylariaceae, Thelephoraceae, and Paxillaceae. Various types of chemical substances were purified and characterized based on the modern spectroscopic methods and also on chemical reactions. These metabolites have shown a broad activity in many biological systems, such as antimicrobial, nematicidal, inhibition of NO production, antihuman immunodeficiency virus, tumor necrosis factor-α, and antioxidant activities. These isolated metabolites did not only show interesting activities, but also are employed as chemical markers supported for chemosystematics of these families. This review paper deals with the chemical constituents of 22 species, their biological activities, and also a discussion on chemosystematics.

Twenty isolates of white rot fungi were collected from several provinces in Thailand. Eight isolates could be cultured to test for thermotolerance and to screen for the presence of ligninolytic enzymes using Phanerochaete chrysosporium as a reference. Of the eight isolates, only three species Daedaleopsis sp., Schizophyllum commune PT, and S. commune SL were able to grow above 40°C. All three species exhibited 2,2 -azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS)-oxidizing activity on ABTS agar plates. Only P. chrysosporium could oxidize manganese on MnCl 2 agar plates. Both P. chrysosporium and Daedaleopsis sp. were able to decolourize wastewater on wastewater agar plates. All fungal cell suspensions tested decolourized wastewater No. 1 (pH 8.07, chemical oxygen demand (COD) 4347 mg/l) from the pulping process and wastewater No. 2 (pH 6.94, COD 4000 mg/l) from the pulping process combined with that from the paper recycling process. Daedaleopsis sp. and P. chrysosporium exhibited the highest ability to decolourize wastewater No. 1 (52%) and No. 2 (86%), respectively. Laccase activities were detected in the decolourized effluents and all fungi tested reduced the COD by 59-71% (No. 1) and 66-83% (No. 2).

Several hundred (396) compounds from New Zealand flora with medicinal properties were analyzed for their physicochemical properties. It was found that approximately 10 % fulfilled all the requirements to be considered to be lead-like, over half of the compounds were deemed to be in the drug-like space and ≈75 % were in the known drug space. These results indicate the presence of a significant proportion of compounds that are of particular interest to pursue as potential lead compounds or therapeutics. Additionally, compound classes were analyzed separately-most carbonyl-containing compounds (aldehydes, ketones, esters and lactones), along with phenols were the most lead-like compounds, which also displayed very good proportions in the drug-like and known drug space. The information presented herein can be harnessed and utilized in future work, through focussing on the compounds and compound classes that exhibit high-levels of lead-likeness for further development.
