Constituents of the Rhizomes of Boesenbergia pandurata and Their Antiausterity Activities against the PANC-1 Human Pancreatic Cancer Line

Abstract: Human pancreatic cancer cell lines have a remarkable tolerance to nutrition starvation, which enables them to survive under a tumor microenvironment. The search for agents that preferentially inhibit the survival of cancer cells under low nutrient conditions represents a novel antiausterity strategy in anticancer drug discovery. In this investigation, a methanol extract of the rhizomes of Boesenbergia pandurata showed potent preferential cytotoxicity against PANC-1 human pancreatic cancer cells under nutrient-… Show more

“…7) Panduratin A and its derivatives have been shown to exert various biological effects, including anti-tumor, anti-viral, and anti-inflammatory activities. [7][8][9] We previously reported the isolation of eight new cyclohexene chalcones and seven known related compounds, e.g., panduratin A, 4-hydroxypanduratin A (Fig. 1A), and isopanduratin A (Fig.…”

“…Given the marked survival ability of tumor cells, anti-austerity agents, compounds that cancel the ability of tumor cells to tolerate nutrient deficiency, are of significant interest [ 9 , 10 , 11 , 12 ]. Wortmannin and chloroquine, autophagy inhibitors, activate caspase-dependent apoptosis in starved PANC-1 cells [ 35 ].…”

“…It is conceivable that compounds that selectively show cytotoxicity against starved cells in tumor microenvironment, i.e., anti-austerity agents, represent potential chemotherapeutics for the treatment and cure of cancer [ 9 ]. Importantly, natural products have been a rich source of anti-austerity agents [ 10 , 11 , 12 ].…”

“…The antiausterity approach 44,45 aims at the identication and development of new targeted agents 44,[49][50][51] with preferential cytotoxicity to pancreatic tumor cells under nutrient-decient conditions. The preferential cytotoxicites are represented as PC 50 values, i.e., the concentration at which 50% of the pancreatic cancer cells were killed in NDM without exhibiting toxicity at normal nutrient-rich medium.…”
