Construction and Aromatization of Hantzsch 1,4‐Dihydropyridines under Microwave Irradiation: A Green Approach

Abstract: Hantzsch reaction, leading to the synthesis of 1,4-dihydropyridines (1,4-DHPs) with momentous biological activities and pharmacological properties is a well-known MCR. In recent years, microwave irradiation (MWI) has been successfully used as the unconventional and green source of energy to promote organic transformations including Hantzsch 1,4-dihydropyri-dines. Literature survey disclosed that construction of 1,4-DHPs as well as their aromatization can be successfully performed under MWI in solution as well … Show more

“…In this study, the synthesis of new 1,4‐dihydropyridine derivatives with methyl or ethyl esters in positions 3 and 5 and the substitution of different ethers in the C2 position of the dihydropyridine ring were performed ( 5 a – h ). In the first step, 1,4 dihydropyridine was synthesized using Hantzsch's synthesis [21] . As a result, symmetric 1,4‐dihydropyridines ( 2 a – b ) were prepared from thiophene‐2‐carbaldehyde and ammonium acetate and β‐Keto esters [22] .…”

“… 3 Usually, the synthesis of PHQ derivatives involves the coupling of three or four components, such as dimedone, aldehyde, ethyl acetoacetate, and ammonia reflux in ethanol or acetic acid. 4 In drug discovery, the multi-component reaction plays a pivotal role. Therefore, for synthetic purposes, multi-component reactions are a more favored approach by younger researchers in academic and industrial realms.…”
