Contact activation in shock caused by invasive group A Streptococcus pyogenes

Abstract: Prolongation of activated partial thromboplastin time in streptococcal toxic shock syndrome is associated with activation of the contact system, possibly contributing to the profound shock associated with streptococcal toxic shock syndrome.

“…Mice showed systemic signs of sickness 8 to 12 hours after infection. To measure clotting times, 8 groups of female BALB/c mice were infected; and after various time points (0,4,6,10,12,18,24, and 42 hours), the animals were anesthetized with isofluorane and terminal blood samples were taken. Approximately 0.5 mL of blood was drawn by cardiac puncture into polypropylene tubes containing one-tenth volume of 3.8% trisodium citrate.…”

“…Contact activation in vivo and treatment with HKH20. Mice were injected subcutaneously in the neck with 2 ϫ 10 7 CFU S pyogenes bacteria, plasma was collected at 0, 4,6,10,12,18,24, and 42 hours after infection (n ϭ 2-6/group), and aPTT (A) and PT (B) were measured immediately. *P Ͻ .05; ***P Ͻ .001.…”

“…However, various reports of decreasing profit in dairy farming have been identified where improvements can be made (Bishop, 1964). Of them, postpartum uterine infection is the majorcause of economic loss in dairy cows (Fox et al, 2002;Sriskandan et al, 2000). It may be due to a longer calving interval, costs of extra services, treatment and increased culling rate (Drillich et al, 2005;Gilbert et al, 2005).…”

“…In addition, the surface-bound M protein, one of the classical virulence determinants of GAS, has been demonstrated to bind HK, followed by bradykinin generation (9,10). Binding and assembly of contact system factors on the bacterial surface are thought to be sufficient to activate this proteolytic system (6); however, additional mechanisms must exist, since Sriskandan et al documented that streptococcal culture supernatants were able to activate PK in vitro (11). Many pathogenic streptococci secrete streptokinase in order to accelerate the conversion of human plasminogen to plasmin (12).…”
