Continuous mixing of powder mixtures with pharmaceutical process constraints

Abstract: While it would provide many advantages from many aspects, the application of continuous mixing processes to the pharmaceutical field is still in its infancy. In this paper we report results concerning the continuous mixing of nine ingredients (including three actives) that constitute a current drug. We examine these results in the light of different pharmaceutical process constraints, such as mixture quality control, time-stability of this quality, sensitivity of the process to perturbations. The apparatus is … Show more

“…In Equation (32) , is the residence time distribution at position x. is the variance at position x and can be calculated as in Equation (33) where is the variance decay rate for a continuous mixing process (33) This allows continuous mixing processes to be readily compared with batch processes. The relationship between the batch and continuous mixing efficiency can be defined in terms of the variance decay rate as in Equation (34).…”

“…[117,118] Optimization of a mixing process often faces constraints as qualification of the characteristics of powder particles requires lengthy and multi-step operations. [116] This is particularly marked when the powder mixing involves a wide variety of excipients, in addition to drug.…”

“…The mixture quality of powder has been a common problem faced by industrialists. [116] The quality of polymer powder mixing is dependent on size, density, geometry, moisture content, surface texture and electrostatic charging of the solid particles. [117,118] Optimization of a mixing process often faces constraints as qualification of the characteristics of powder particles requires lengthy and multi-step operations.…”

“…In one of our works [6], we examined the effect of process disturbance, such as that occurring during periods of feeding of loss-in-weight feeders. In particular, we quantified the response of the mixer to filling sequences of two critical feeders in terms of mixture homogeneity.…”
