Control of gallbladder contractions by cholecystokinin through cholecystokinin-A receptors on gallbladder interstitial cells of cajal

Abstract: AIM:To identify the cholecystokinin (CCK)-A receptors (CCK-AR) on the guniea pig gallbladder interstitial cells of cajal (ICC) and to study CCK-8 induced gallbladder muscle strip contractions through the CCK-AR. METHODS:The existence of CCK-AR was examined by immunohistofluorescence on sectioned tissue and cultured cells. In vitro contractile response of guinea pig gallbladder muscle strips and the strips with ICC removed were also studied with CCK-8 receptors added. RESULTS:In t i s s ue s e c t ions , int e … Show more

“…Rats were purchased from the Experiment Animal Center of Tongji Medical College, Huazhong University of Science and Technology, Wuhan, China. They were housed under normal laboratory conditions (12/12-hour light-dark cycle, 18 C-22 C) and were given water ad libitum. The study was started after 1 week, when the rats were adapted to the environment.…”

“…ICLCs are believed to act as sources of spontaneous, electric slow waves responsible for paced contractions of GBSM, thus regulating the rhythm of gallbladder bile release. 27,28,30 A significantly smaller number of ICLCs was found in gallbladders from patients with cholelithiasis compared with those without gallstones. 31 Furthermore, a negative correlation between the number of ICLCs and bile CSI was discovered.…”
