Controlled release of recombinant human cementum protein 1 from electrospun multiphasic scaffold for cementum regeneration

Abstract: Periodontitis is a major cause for tooth loss, which affects about 15% of the adult population. Cementum regeneration has been the crux of constructing the periodontal complex. Cementum protein 1 (CEMP1) is a cementum-specific protein that can induce cementogenic differentiation. In this study, poly(ethylene glycol) (PEG)-stabilized amorphous calcium phosphate (ACP) nanoparticles were prepared by wet-chemical method and then loaded with recombinant human CEMP1 (rhCEMP1) for controlled release. An electrospun m… Show more

“…The interaction between BSA and metallic cations also resulted in a downshift of amide band I signal at 1646-1633 cm −1 as per HSAB Lewis's theory of binding (Bronze-Uhle et al, 2016). The formation of BSA-NF also led to the crystallization of phosphate ions from PBS and this was confirmed by the appearance of new peaks at 1033 and 561 cm −1 (Sun et al, 2016) (Figure S1a).…”

“…The application of this protein as an agent promoting cementum regeneration would also be interesting. The effects of bioresorbable scaffold loaded with rhCEMP-1 on the attachment, proliferation, and osteogenic and cementogenic differentiations of human periodontal ligament cells have been investigated histologically in a critical size defect on rodent, and the potential of generating cementum-like tissue in vitro and in vivo has been demonstrated [21].…”

“…Several approaches have been used and tested to regenerate teeth [ 67 ]; these approaches have focused on targeted regeneration of individual tooth components such as dental pulp and dentin [for reviews see 68 – 70 ; for functional studies see 70 73 ], cementum [for reviews see 74 , 75 ; for functional studies see 76 78 ], periodontium [for reviews see 79 , 80 ; for functional studies see 81 83 ], and enamel [for reviews see 84 ; for functional studies see 85 88 ]. However, combining these strategies does not guarantee the successful regeneration of a viable tooth.…”
