Corneal Preparation of Eye Bank Eyes for Experimental Surgery

Abstract: A solution of 20% dextran in BSS is effective for dehydrating eye bank corneas to a quasi-normal physiologic thickness and for maintaining the thickness for a sufficient amount of time. Both the posterior and anterior sides of the cornea should be exposed to the solution for 60 minutes.

“…Follow-up protected t tests revealed that after the application of isoosmolar riboflavin solution with dextran for 30 minutes, the thinnest pachymetric measurements decreased significantly by 55. 22 Intraoperative Pachymetry in CCL in Thin Corneas hypoosmolar riboflavin application for 10 minutes compared with thickness at the end of isoosmolar riboflavin application (P , 0.01). However, this artificial swelling effect was transient, and the thinnest pachymetric readings decreased significantly after 10 and 30 minutes of isoosmolar riboflavin application compared with the thickness at the end of hypoosmolar riboflavin application (P , 0.01 for both).…”

“…In addition to cytotoxicityrelated problems, CXL can also be expected to have less effect on the biomechanics of an artificially swollen cornea because of the lower relative concentration of collagen in the hydrated stroma, as has been reported with collagen gels. [19][20][21] Some studies also used hypoosmolar riboflavin solution before the irradiation and standard dextran-riboflavin solution Means G SD *After 30 minutes of riboflavin during the actual irradiation, 14 which is critical because of the possible reduction of the previous swelling induced by the dextran solution due to the hyperosmolar effect of the 20% dextran. 19 An alternative for thin corneas is the C3-R technique, in which the epithelium is not removed and preoperative application of benzalkonium chloride drops loosens the tight junctions of the epithelium.…”

“…The 20% dextran solution was prepared by dissolving 20 g of 229,000 MW dextran (Sigma, St Louis, MO) in 100 ml balanced salt solution (BSS, Alcon, Forth Worth, TX), as done and proposed in previous studies [5,13,18,26,27]. The above-mentioned procedure for data acquisition was then carried out in the same manner immediately thereafter.…”
