The use of entomological specimens as a source of forensic DNA evidence has been studied since the late 20 th century. Currently as it stands, to preserve both the minimum postmortem interval (mPMI) and DNA evidence within fly larvae, two standards of preservation exist. For mPMI, the use of hot water kill with near-boiling water is used to fix larvae size for accurate age estimation. For DNA evidence, the standard technique of preservation is submersion in 70% ethanol. To help reduce the number of standard preservation techniques for practitioners, this study assessed the feasibility of the hot water kill preservation technique using near-boiling water when obtaining short tandem repeat (STR) profiles from the contents of fly larvae crops (foregut). Reduction in the preservation techniques would result in the ability to collect only one set of specimens that could be used for both mPMI determination and DNA collection.

The designing of Computer-Aided Diagnosis (CADx) is necessary to improve patient condition analysis and reduce human error. HistAENN (Histogram-based Autoencoder Neural Network, the first hierarchy level) and the fractal-based estimator (the second hierarchy level) are assumed for segmentation and image analysis, respectively. The aim of the study is to investigate how to select or preselect algorithms at the second hierarchy level algorithm using small data sets and the semisupervised training principle. Method-induced errors are evaluated using the Monte Carlo test and an overlapping table is proposed for the rejection or tentative acceptance of particular segmentation and fractal analysis algorithms. This study uses lung histological slides and the results show that 2D box-counting substantially outweighs lacunarity for considered configurations. These findings also suggest that the proposed method is applicable for further designing of classification algorithms, which is essential for researchers, software developers, and forensic pathologist communities.

Histological quantification of decomposed human livers: a potential aid for estimation of the post-mortem interval?

The objective of this study was to determine if a novel scoring-based model for histological quantification of decomposed human livers could improve the precision of post-mortem interval (PMI) estimation for bodies from an indoor setting. The hepatic decomposition score (HDS) system created consists of five liver scores (HDS markers): cell nuclei and cell structure of hepatocytes, bile ducts, portal triad, and architecture. A total of 236 forensic autopsy cases were divided into a training dataset (n = 158) and a validation dataset (n = 78). All cases were also scored using the total body score (TBS) method. We specified a stochastic relationship between the log-transformed accumulated degree-days (log10ADD) and the taphonomic findings, using a multivariate regression model to compute the likelihood function. Three models were applied, based on (i) five HDS markers, (ii) three partial body scores (head, trunk, limbs), or (iii) a combination of the two. The predicted log10ADD was compared with the true log10ADD for each case. The fitted models performed equally well in the training dataset and the validation dataset. The model comprising both scoring methods had somewhat better precision than either method separately. Our results indicated that the HDS system was statistically robust. Combining the HDS markers with the partial body scores resulted in a better representation of the decomposition process and might improve PMI estimation of decomposed human remains.
