Correlative biochemical and morphological studies of myelination in human ontogenesis

Abstract: Biochemical and morphological observations of nerve roots in six fetuses from the 16th to 34th week of gestation and five infants 1 day to 3 years old are presented. In dorsal roots the process of myelination begins later than in the ventral roots and spinal cord and proceeds much slower. As in the spinal cord during nerve roots myelin maturation profound lipid changes are observed.

“…Axons of this morphology were very rarely seen in control VRs, even as early as 1 day of life (fig. S3, R and T), but are well documented in human fetal nerve (21,22). This preponderance of directly abutting axons was also found in SMA intercostal nerves (fig.…”

“…2G). Myelination occurs earlier in motor than sensory roots (Niebroj-Dobosz et al, 1980), and expression of myelin-specific mRNAs is detectable at P1 in motor but not sensory roots (Baron et al, 1994;Forghani et al, 2001). Thus, to correlate the onset of DG with the beginning of myelination, we stained motor and sensory roots at P1.…”

“…Literature data show that myelination usually begins when axons have reached a caliber of about 1 m, so that those Þbers that are the Þrst to be myelinated correspond to the largest Þbers in mature nerves [24]. Moreover, myelination of motor roots is known to precede that of sensory ones [32]. Therefore, nerve Þbers which Þrst undergo myelination and mostly represent motor axons could have been mainly affected by prenatal CO exposure.…”
