Creating a 3D microenvironment for monocyte cultivation: ECM-mimicking hydrogels based on gelatine and hyaluronic acid derivatives

Abstract: A co-crosslinked gelatin and hyaluronan based matrix is a suitable microenvironment for the 3D culture of THP-1 cells, which enhances their differentiation into macrophages.

“…Thus, it has been used in many studies. [42][43][44][45][46][47] Biocompatibility was assessed by incubating cells on the surface and inside of Gel-Ty hydrogel. Cell viability was not significantly different between hydrogels at all concentrations.…”

“…In our previous study, we have demonstrated that the HRP-mediated crosslinking reaction of HA-TA-and GTN-HPA-based polymers is a highly tunable system where the degree of crosslinking and therefore the stiffness of the hydrogel can be adjusted by the concentration of hydrogen peroxide and activity of HRP. 36 To achieve comparable mechanical strength between hydrogels containing different ratios of HA-TA and GTN-HPA, decreased concentration of the crosslinking agents was used in the case of the HA-TA hydrogel. The prepared HA-TA + GTN-HPA hydrogels had only slightly higher G 0 values as compared to the GTN-HPA hydrogels; the other mechanical properties were comparable among all the three materials.…”

“…Hydrogel preparation was described in our previous study. 36 Briey, the HA-TA derivative was dissolved in a mixture of 0.9% NaCl + RPMI (1 : 1) at RT overnight, and the GTN-HPA conjugate was dissolved in the HA-TA solution at 37 C for next 2 h. The two-compound system was used for hydrogel/coating preparation. The solution (HA-TA or HA-TA + GTN-HPA (1 : 10 w/w) or GTN-HPA) was equally divided into two parts.…”

“…Fortunately, recent investigations on biomacromolecules have employed light-scattering devices 31 . That proves advantageous because light-scattering is an extremely sensitive technique for measuring absolute Mw 32 . For example, Ricci et al investigated the influence of Mw of HA by light-scattering device on the nanovector properties 33 ; Botha et al analyzed the Mw of hydrophobically modified HA by size-exclusion chromatography and light-scattering combination method 34 .…”
