Critical link between TRAIL and CCL20 for the activation of TH2 cells and the expression of allergic airway disease

Abstract: The role of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) in immune responses mediated by T-helper 2 (T(H)2) lymphocytes is unknown. Here we characterize the development of allergic airway disease in TRAIL-deficient (Tnfsf10(-/-)) mice and in mice exposed to short interfering RNA targeting TRAIL. We show that TRAIL is abundantly expressed in the airway epithelium of allergic mice and that inhibition of signaling impairs production of the chemokine CCL20 and homing of myeloid dendritic cells a… Show more

“…Since DR5 agonistic antibody treatment was more similar to DR5 engagement with TRAIL, our results suggest that TRAIL might be able to induce inflammatory cytokine release under certain circumstances. Interestingly, Weckmann et al [33] reported a link between TRAIL and CCL20, which supports our conclusion.…”

“…AHR, eosinophil migration, and mucus production are regulated by the T H 2 cytokines IL-5 and IL-13 (26)(27)(28)(29)(30), and production of these cytokines were inhibited by ant-miR-126 treatment. Antagomir treatment also inhibited the recruitment of CD4 T cells (predominately T H 2 cells in this model) and T H 2 promoting myeloid DCs into the airways, which are critical for the expression of allergic airways disease (39)(40)(41). Importantly, overall suppression of the T H 2 response by ant-miR-126 occurred without evidence of a switch to a T H 1 response, as T cell IFN␥-secretion and neutrophil recruitment were not promoted by antagomir treatment.…”

“…Another difference between the two is that post-CD3 stimulation, Thl cells will up-regulate CD95L, where Th2 cells express TRAIL. TRAIL regulates more than apoptosis, with gene disruption leading to inhibition of DC and Th2 cells to allergy sites in mice (Weckmann et al, 2007). This led to reduction in Th2 cytokine release and a subsequent release in inflammation.…”
