Croatian Society of Medical Biochemistry and Laboratory Medicine: National recommendations for blood collection, processing, performance and reporting of results for coagulation screening assays prothrombin time, activated partial thromboplastin time

Abstract: A modern diagnostic laboratory offers wide spectrum of coagulation assays utilized in the diagnosis and management of patients with haemostatic disorders, preoperative screening and anticoagulation therapy monitoring. The recent survey conducted among Croatian medical biochemistry and transfusion laboratories showed the existence of different practice policies in particular phases of laboratory process during coagulation testing and highlighted areas that need improvement. Lack of assay standardization togethe… Show more

“…Coagulation biomarkers (prothrombin time (PT), activated partial thromboplastin time (aPTT), fibrinogen (Fbg), D-dimers)) were measured on BCS XP coagulation analyser (Siemens, Marburg, Germany), by the following methods: PT – photo-optical method (Dade Innovin %), APTT – photo-optical method (Dade Actin FS), Fbg – modification of the Clauss method (Multifibren U), and D-dimers – particle-enhanced immunoturbidimetric assay (Innovance D-dimer)). An aPTT-ratio was calculated according to the National recommendations for blood collection, processing, performance and reporting of results for coagulation screening assays prothrombin time, activated partial thromboplastin time, thrombin time, fibrinogen and D-dimer ( 19 ). The recommendations state that the aPTT ratio is calculated as the ratio of patient’s aPTT divided by the mean value of the reference interval for the particular system (reagent/coagulometer) used.…”

“…CBC was performed within half an hour from blood sampling. Tubes with sodium citrate were centrifuged two times at 1500 x g for 15 min, as per national recommendations [33], and serum tubes with gel were centrifuged at 2000 g for 10 min, as recommended by the manufacturer. WBC counts were performed on a Sysmex XN-1000 analyzer (Sysmex Corporation, Kobe, Japan) as part of the CBC.…”

“…If samples were stored frozen for a certain period of time, the thawing procedure may affect sample integrity, at least for some analytes (e.g. coagulation parameters) [17]. For this reason, if samples were stored frozen, authors should provide the detailed description about how samples were thawed and the number of freeze thaw cycles.…”
