Crystal structure determination and site-directed mutagenesis of the Pyrococcus abyssi aCBF5-aNOP10 complex reveal crucial roles of the C-terminal domains of both proteins in H/ACA sRNP activity

Abstract: In archaeal rRNAs, the isomerization of uridine into pseudouridine (Ψ) is achieved by the H/ACA sRNPs and the minimal set of proteins required for RNA:Ψ-synthase activity is the aCBF5–aNOP10 protein pair. The crystal structure of the aCBF5–aNOP10 heterodimer from Pyrococcus abyssi was solved at 2.1 Å resolution. In this structure, protein aNOP10 has an extended shape, with a zinc-binding motif at the N-terminus and an α-helix at the C-terminus. Both motifs contact the aCBF5 catalytic domain. Although less effi… Show more

“…To address this question, a great deal of efforts have been expended to solve the crystal structures of archaeal box H/ACA snRNP complexes. As a result, the structures of various forms, from the initial complex of three core proteins to a complete, substrate-bound box H/ACA RNP, have been solved (Li and Ye, 2006;Manival et al, 2006;Rashid et al, 2006;Liang et al, 2007;Duan et al, 2009;Liang et al, 2009). A detailed picture of how this most complex pseudouridylase modifies its substrate has now become available.…”

“…The structure of the archaeal pseudouridylase Cbf5 was solved complexed with Nop10 [5,6] or with Nop10 and Gar1 [4]. Based on structural and sequence homologies, pseudouridylases are classified into 5 families, RluA, RsuA, TruA, TruB, and TruD [10].…”

“…Recently, three groups solved the crystal structures of archaeal H/ACA RNP core complexes consisting of two or three core proteins (Fig. 1) [4][5][6]. For the first time, these structures provide molecular details of protein-protein interactions in the H/ACA core complex and of the pseudouridylase itself.…”

“…Conversely, the exchange of the other core proteins could reflect a possible rearrangement or breathing of the complex during catalysis. For example, recent crystal structures of partial archaeal H/ACA protein complexes demonstrate how NOP10 brackets the catalytic domain of NAP57 (Hamma et al 2005;Manival et al 2006;Rashid et al 2006). During catalysis this NOP10-NAP57 interaction may need to be loosened to allow access to and/or release of substrate RNAs.…”
