Crystal structure of the solute-binding protein BxlE from Streptomyces thermoviolaceus OPC-520 complexed with xylobiose

Abstract: BxlE from Streptomyces thermoviolaceus OPC-520 is a xylo-oligosaccharide (mainly xylobiose)-binding protein that serves as the initial receptor for the bacterial ABC-type xylo-oligosaccharide transport system. To determine the ligand-binding mechanism of BxlE, X-ray structures of ligand-free (open form) and ligand (xylobiose)-bound (closed form) BxlE were determined at 1.85 Å resolution. BxlE consists of two globular domains that are linked by two β-strands, with the cleft at the interface of the two domains c… Show more

“…The DALI server indicated that the NagB1/(GlcNAc) 2 structure was similar to those of various sugar SBPs such as the glucose binding protein (PDB: 5DVI) of Pseudomonas putida CSV86 ( Pandey et al, 2016 ), with a Z-score of 40.3 (r.m.s.d. for the 396 Cα atoms = 2.5 Å and an a.a. sequence identity = 19%), xylooligosaccharide-binding protein BxlE (PDB: 3VXB) from S. thermoviolaceus OPC-520 ( Tomoo et al, 2017 ) with a Z-score of 38.3 (r.m.s.d. = 4.3 Å (399 atoms) and a.a. sequence identity = 24%), and raffinose binding protein RafE (PDB: 6PRE) of Streptococcus pneumoniae ( Hobbs et al, 2019 ) with a Z-score of 38.0 (r.m.s.d.…”

“…Thaxtomin itself, instead of the cellulolytic system, would be responsible for generating the inducer of its own biosynthesis, as previously proposed by Johnson et al This hypothesis implies that S. scabies would have evolved a mechanism able to monitor very low concentrations of cellotriose emanating from root exudates. This seems to be the case, as we have shown that CebE of S. scabies has higher binding affinity for the trisaccharide (dissociation constant [ K d ] for cellotriose, ~2 nM) than for the disaccharide ( K d for cellobiose, ~14 nM), which is unusual for Streptomyces ABC-type sugar-binding components ( 12 , 15 , 16 ). Whether the presence of cellotriose and the absence of cellobiose are constants in exudates from roots and tubers commonly damaged by S. scabies remains to be demonstrated.…”
