Culture media-based selection of endothelial cells, pericytes, and perivascular-resident macrophage-like melanocytes from the young mouse vestibular system

Abstract: The vestibular blood-labyrinth barrier (BLB) is comprised of perivascular-resident macrophage-like melanocytes (PVM/Ms) and pericytes (PCs), in addition to endothelial cells (ECs) and basement membrane (BM), and bears strong resemblance to the cochlear BLB in the stria vascularis. Over the past few decades, in vitro cell-based models have been widely used in blood-brain barrier (BBB) and blood-retina barrier (BRB) research, and have proved to be powerful tools for studying cell-cell interactions in their respe… Show more

“…Similarly, PVMs in peripheral organs restrict vascular permeability, such as mesentery arteries and cochlear . Comparable finding is also reported in an in vitro vestibular BBB co‐culture system consisting of endothelial cells (ECs), PVMs, and pericytes . Interestingly, PVMs may lead to BBB disruption in diseased status.…”

“…Culture media‐based selection and growth of primary cells is based on specific culture media, formulated to selectively grow certain cell phenotypes, when starting from a specific cell source. Such methods were proven to be very reliable, providing consistent results (Zhang et al, ). To the best of our knowledge, the current study is the first describing a protocol for culturing strial PC from bovine cochlea.…”

“…Strial melanocytes in cochlea and perivascular‐resident macrophage‐like melanocytes (PVM/Ms) in vestibular end organ had been described by several researchers (Escobar, Zuasti, Ferrer, & Garcia‐Ortega, ; Murillo‐Cuesta et al, ; Tachibana, ; Zhang et al, ). Their crucial role in the production of evoked potentials via Na + K+‐ATPase and potassium channels and maintenance of the blood‐labyrinth barrier are the main functions assigned to the melanocytes in the labyrinth.…”
