Current Approaches in Antiviral Drug Discovery Against the Flaviviridae Family

Abstract: Viruses belonging to the Flaviviridae family primarily spread through arthropod vectors, and are the major causes of illness and death around the globe. The Flaviviridae family consists of 3 genera which include the Flavivirus genus (type species, yellow fever virus) as the largest genus, the Hepacivirus (type species, hepatitis C virus) and the Pestivirus (type species, bovine virus diarrhea). The flaviviruses (Flavivirus genus) are small RNA viruses transmitted by mosquitoes and ticks that take over host cel… Show more

“…Three antiviral candidate therapies, including ribavirin, interferon α (IFN-α), and WNV-specific immunoglobulin are discussed below. Emerging therapies that lack human data, including nucleic acid inhibition, peptides, and small molecules targeting viral translocation, replication, or protease activity, have been reviewed recently and are not discussed here 87,88…”

“…At the moment, patients with severe cases of Flavivirus infections are treated only by supportive care, which includes intravenous fluids, hospitalization, respiratory support, and prevention of secondary infections. [14] In fact, despite worldwide extensive research and public health concern associated with Flavivirus diseases, no specific/routine treatment is available today to combat any Flavivirus infections, the sole exception being constituted by new drugs (e.g., simeprevir and sofosbuvir) currently in clinical trials for hepatitis C. [15,16] Consequently, there is an urgent need for new lead compounds that efficiently target distinct stages of the viral replication cycle in a virus-specific way.…”

“…Considering the global threat of flaviviral pandemics, specific treatments are craved for. 44 3.1.1 ZIKA virus ZIKV is transmitted to humans by mosquito bites: in adults, it usually causes mild to no symptoms, while it might be devastating if contracted during pregnancy, as it is related to microcephaly in the unborn child. Several G4 forming sequences have been discovered in the positive strand of the ZIKV genome, while the negative one is devoid of PQSs.…”
