Cutting the ovarian surface improves the responsiveness to exogenous hormonal treatment in aged mice

Abstract: Fertility decreases with increasing age, and natural pregnancy becomes difficult in most women over 40 years of age. 1,2 Infertility care using assisted reproductive technology (ART) with control ovarian stimulation (COS) supports women who desire pregnancy. 3,4 COS is an exogenous FSH/hMG treatment method to collect a large number of mature oocytes that are used for in vitro fertilization (IVF). 5 However, the number of collected oocytes is limited in some patients who receive COS treatment, and these cases a… Show more

“…PCOS is a common endocrine disorder among women of reproductive age, and it is associated with reproductive disorders, such as infertility and anovulation-related menstrual disorders, and metabolic disturbances. [1][2][3]19,20 As noted above, various kinds of animal and/or uterine weights. 11,13,21 Similarly, the DHT-induced PCOS animal model used by Osuka et al had small ovaries without cystic morphological ovarian changes.…”

“…PCOS is a common endocrine disorder among women of reproductive age, and it is associated with reproductive disorders, such as infertility and anovulation‐related menstrual disorders, and metabolic disturbances. 1 , 2 , 3 , 19 , 20 As noted above, various kinds of animal models have been used to evaluate the endocrine, metabolic, and histological effects of PCOS on tissues and organs. 9 , 10 Although DHT‐induced PCOS animal models have frequently been used for reproductive and metabolic evaluations in previous studies, they did not completely reproduce all of the phenotypes of human PCOS.…”

“…Aged patients exhibit decreased ovarian stromal blood flow owing to a marked reduction in the number and caliber of blood vessels with changes in endothelial cells [42]. Insufficient supply of growth factors owing to decreased blood flow causes follicular arrest and ovarian fibrosis in aged women with POR [43,44]. SMinduced changes in FGF signaling could drive tissue repair in aged ovaries pre-or post-ovulation [45,46].…”

“…Abrogating fibrosis using hormonal treatment or mechanical stimuli has been shown to restore follicular development and fertility in a mutant mouse model of precocious ovarian aging [56,57]. In addition, antifibrotic drug treatment eliminates fibrotic collagen and restores ovulation in reproductively aged mice [58].…”
