Cyclops‐independent floor plate differentiation in zebrafish embryos

Abstract: In zebrafish, development of the ventral neural tube depends on the Nodal-related signal Cyclops (Cyc). One-day-old cyc mutant embryos lack the medial floor plate (MFP). We show here that cells expressing MFP marker genes differentiate gradually in cyc mutant embryos in a delayed manner during the second day of development. This late differentiation is restricted to the hindbrain and spinal cord and depends on an intact Hedgehog (Hh) signalling pathway. Cells expressing MFP marker genes in cyc mutant embryos a… Show more

“…In Nodal 2/2 mutants, the floor plate is prematurely induced in the anterior epiblast in mouse embryos (Camus et al 2006), whereas inactivation of the gene encoding the nodal antagonist Lefty increases induction of mesendoderm in formerly ectodermal tissue (Meno et al 1999;Perea-Gomez et al 2002). Nodal-deficient mutants show late recovery of midline floor plate cells in the trunk and tail, identifying signaling pathways that ameliorate these effects, specifically Shh and Gli activity (Albert et al 2003;Norton et al 2005). This late recovery, however, is not observed in the brain.…”

“…2AЉ). Therefore, although Hh signalling is required to maintain medial floor plate (Albert et al, 2003), it is not required for the initial formation of this structure.…”

“…1). We used transgenic embryos that express the green fluorescent protein (gfp) gene under control of the regulatory elements of the shh gene (Albert et al 2003). At the 6-10-somite stage, these transgenic embryos express the gfp reporter gene in the notochord, the prechordal plate, and in the ventral midline of the neural plate comprising the prospective floor plate and hypothalamus ( Fig.…”

“…For each locus, the coding region with the intronic sequences, 10 kb upstream of the ATG and 10 kb downstream from the stop codon, were searched for homologies with the "Compare" and "DotPlot" programs (GCG Wisconsin package). The settings of the search algorithm were kept at the default values, which allowed scoring the regulatory sequences of the zebrafish ngn1 and shh genes (Muller et al 1999(Muller et al , 2000Albert et al 2003). Among 45 pairwise comparisons, 18 homologies were detected in the noncoding regions of 10 genes ( Fig.…”
