Cyclosporin‐Induced Gingival Overgrowth at the Newly Formed Edentulous Ridge in Rats: A Morphological and Histometric Evaluation

Abstract: An enlarged edentulous ridge and an altered dental alveolar bone morphology were observed in CsA-treated animals at the end of the study; therefore, we suggest that CsA may induce not only a soft tissue overgrowth but also an alveolar bone alteration at the edentulous ridge. The hypothesis that tooth or periodontal ligament is an essential component for the overgrowth development is questioned.

“…We recently reported similar findings of an enlarged gingival enlargement and associated osteoporotic bony changes in the newly formed edentulous ridge 9 . In our previous study, however, we allowed the ridge to heal for the first two weeks after tooth extraction and then administered CSA at a dose of 15 mg/kg by gastric feeding for four weeks.…”

“…The histological preparation and measurement of the soft and hard tissues around the maxillary dental alveolus were performed following a technique described in our previous studies 9,12,13 . In brief, the right maxillae were dissected and fixed in 70% ethanol and embedded in methyl-methacrylate.…”

“…In vivo studies indicate that CsA accelerates bone remodeling and results in bone loss. 18 In CsA-treated rabbits, a significant decrease in the bone area next to the implant has been recorded, whereas the degree of bone-to-implant contact observed was found to be comparable in test and control groups. 16 Increased bone remodeling and trabecular bone loss have been observed in CsAexposed animals.…”

“…These 2 studies were designed to assess the influence of CsA administration during bone healing immediately after placement of dental implants. Other investigations have reported gingival hyperplasia 8,18 and periodontal damage. 19 Ozone (O 3 ) has a range of valuable properties that can be exploited for application in medical fields.…”

“…A variety of direct and/or indirect interactions between CsA and gingival fibroblasts have been investigated, including metabolic and synthetic activities 4‐8 . However, studies on the effects of CsA on gingival epithelial cells are limited 9‐11 . Although CsA stimulates the production of keratinocyte growth factor, 12,13 which may cause epithelial proliferation in overgrown gingiva, exactly how the drug elicits epithelial hyperplasia in the gingiva is still under investigation.…”
