Cytochemical characterization of sulfo- and sialoglycoconjugates of human laryngeal glandular cells.

Abstract: The composition and distribution of d o -and sialoglyamnjugates in human laryngeal glands have been investigated at light and electron microscopic levels by use of peroxidase-, digoxigenin-, and colloidal gold-conjugated lectins in combination with several chemical and enzymatic deglycosylation procedures. The present study reveals a variety of terminal Oligosaccharide sequences in serous and mucous glands. Serous cells contained glymnjugates with terminal Neu5Ac(a2-3) Gal (81-4) GlcNAc, Neu5Ac (a2-6) GallGalN… Show more

“…In accordance with many authors (Lamb and Reid 1972;Jones and Reid 1978;Jacob and Poddar 1982;Spicer et al 1983;Castells et al 1990Castells et al , 1991Castells et al , 1992Castells et al , 1994 Mandal and Mandal 1990;Jeffery et al 1992), we found both neutral and acidic sulphated and acidic sialylated glycoconjugates in the mucous granules of goblet cells in the tracheal epithelium of the control rabbits. In experimental groups, we identified the same glycoconjugates, too.…”

“…The presence of neutral and acidic sulphated and acidic sialylated glycoconjugates (GCCs) was widely evidenced in many species including humans (Lamb and Reid 1972;Jones and Reid 1978;Jacob and Poddar 1982;Spicer et al 1983;Castells et al 1990Castells et al , 1991Castells et al , 1992Castells et al , 1994Jeffery et al 1992;K im et al 1997). Damjanov (1987) stressed the differences in the GCC composition among individual species.…”

“…The use of desulphation techniques in combination with lectin cytochemistry has allowed the identification of sulphated carbohydrates in the oligosaccharidic chains of glycoproteins Castells et al, 1994). Desulphation methods would also be interesting in studies of gallbladder glycoproteins.…”

“…This WGA labelling pattern reveals the presence of membrane integral proteins associated with these protective complex chains of oligosaccharides, in late endosomes, heterolysosomes and residual bodies. Moreover, the DSA lectin, which is related to the presence of lamps in lysosomal compartments (Castells et al 1994;Egea et al 1989), is localised in the inner membrane of numerous vesicles of the ELS in digestive cells. DSA exhibits an extremely high affinity for Galβ1-4GlcNAc residues (Green et al 1987;Yamashita et al 1987) and, therefore, its labelling distribution confirms the presence of the protective integral proteins in the membrane of the acidic compartments, in agreement with the results obtained with WGA.…”
