Cytokine detection and simultaneous assessment of rheumatoid factor interference in human serum and synovial fluid using high-sensitivity protein arrays on plasmonic gold chips

Abstract: BackgroundFluorescence-enhancing microarray on plasmonic gold film is an attractive alternative to traditional enzyme-linked immunosorbent assay (ELISA) for cytokine detection because of the increased sensitivity. The assay chemistry is similar to an ELISA sandwich assay, but owing to the gold substrate, cytokine measurements are 10 to 100 times more sensitive and can be multiplexed. Plasmonic protein microarrays are, as other immunoassays, affected by the presence of heterophilic antibodies and rheumatoid fac… Show more

“…The concentrated electromagnetic field built around a metal will increase the excitation of the fluorophore. For instance, the detection sensitivity improved with a plasmonic Au chips (Valentina et al, 2015). Other sensitivity‐enhancing techniques include photonic crystals and optical resonance, where shifts in the resonant wavelength can be correlated with antigen (cytokine) binding events at the sensor surface (Maier, Morgan, Lindner, & Pittner, 2008; Mathias, Ganesh, & Cunningham, 2008).…”

“…Further, the expression of IL-20 in RA synovial tissue was shown to be particularly associated with macrophages, neutrophil granulocytes and fibroblast-like synovial cells ( 18 ). IL-19, IL-20, and IL-24 have all been identified in both synovial fluid and plasma of patients with RA and SpA ( 15 , 16 , 18 20 ).…”
