Cytotoxic and Hepatoprotective Effects of Bupleurum flavum Flavonoids on Hepatocellular Carcinoma HEP-G2 Cells

Abstract: Aims:In this study, we aimed at investigating the possible cytotoxic and hepatoprotective effects of crude extract, flavonoid mixture and pure flavonoids isolated from the aerial parts of Bupleurum flavum Forsk. (Apiaceae) native to Bulgaria. Methods: For the first time flavon C-glycoside vicenin-2 was isolated from B. flavum. Its structure was identified by LC-ESI/MS/MS analysis in positive ion mode. Cytotoxic effects of vicenin-2, as well as the known flavonoid narcissin and B. flavum crude extract at concen… Show more

“…In the same way, 6- C -hexosyl-luteolin was confirmed by the diagnostic ions 0,3 X − at m / z 357.061 (39.3%) and 0,2 X − at m / z 327.051 (53.7%); luteolin ([Lu-H] − at m / z 285.041) was deduced from RDA ions 1,3 B − at m / z 133.028 and 1,4 A − at m / z 175.038. Based on the comparison with reference standards, 62 and 63 were unambiguously identified as vicenin-2 [ 32 ] and homoorientin, respectively.…”

“…In a previous study, Compound 2 did not show cytotoxic activity up to 100 µmol/L on the IGROV-1 and OVCAR-3 human ovary cell lines and HCT-116 human colon cancer cell line ( Znati et al, 2014 ). It was also determined that narcissin (25–400 μg/mL) had no significant hepatoprotective activity when tested on HEP-G2 hepatocellular carcinoma cells for 72 h by MTT assay ( Gevrenova et al, 2016 ). In the present study, the IC 50 values of Compound 2 were 137.10 ± 8.62 and 77.78 ± 39.88 μg/mL on the MCF-7 and MDA-MB-231 cell lines, respectively.…”
