Cytotoxic effects of vincristine on tumour subpopulations separated from pulmonary nodules

Abstract: allowed to recover in the absence of VCR for 24h prior to their removal and separation, FSa cell survival increased in each of the elutriated populations. In contrast, while G1 enriched cell populations from in vitro exposed cultures still exhibited a significant reduction in CFE, no such age-specific response was observed for in vivo exposed macroscopic pulmonary nodules. The stathomokinetic effect of VCR on FSa cells was also readily observed in vitro using FMF analysis (e.g., an increase of from 20 to 38% i… Show more

“…As described in earlier reports, the method of centrifugal elutriation has been shown to be effective in separating and synchronizing FSa cells grown as artificial pulmonary metastases [11][12][13]. Following treatment with AMSA and/or radiation, no discernible difference was observed between the cell populations derived from treated and untreated control FSa nodules with respect to either cell yield following the preparation of single cell suspensions or sedimentation profiles following the separation of cells by centrifugal elutriation.…”

“…An in vivo procedure has been developed which allows for the rapid testing and evaluation of the phase-specific therapeutic effectiveness of selected cytotoxic agents on artificial pulmonary metastases [11][12][13]. In particular, parameters such as the sequencing and timing of therapeutic agents need to be characterized.…”
