Yixinyin, the traditional Chinese medicine, has the effects of replenishing righteous qi, and promoting blood circulation to eliminate blood stagnation. It is often used to treat patients with acute myocardial infarction (MI). The purpose of our study is to explore the key components and targets of Yixinyin in the treatment of MI. In this study, we analyzed gene expression data and clinical information from 248 samples of MI patients with the GSE34198, GSE29111 and GSE66360 data sets. By constructing a weighted gene co-expression network, gene modules related to myocardial infarction are obtained. These modules can be mapped in Yixinyin PPI network. By integrating differential genes of healthy/MI and unstable angina/MI, key targets of Yixinyin for the treatment of myocardial infarction were screened. We validated the key objectives with external data sets. GSEA analysis is used to identify the biological processes involved in key targets. Through molecular docking screening, active components that can combine with key targets in Yixinyin were obtained. In the treatment of myocardial infarction, we have obtained key targets of Yixinyin, which are ALDH2, C5AR1, FOS, IL1B, TLR2, TXNRD1. External data sets prove that they behave differently in the healthy and MI (P < 0.05). GSEA enrichment analysis revealed that they are mainly involved in pathways associated with myocardial infarction, such as viral myocarditis, VEGF signaling pathway and type I diabetes mellitus. The docking results showed that the components that can be combined with key targets in YixinYin are Supraene, Prostaglandin B1, isomucronulatol-7,2′-di-O-glucosiole, angusifolin B, Linolenic acid ethyl ester, and Mandenol. For that matter, they may be active ingredients of Yixinyin in treating MI. These findings provide a basis for the preliminary research of myocardial infarction therapy in traditional Chinese medicine and provide ideas for the design of related drugs.

Background: This systematic review and meta-analysis aimed to evaluate the effects of Taohong Siwu Decoction (THSWD) combined with low molecular weight heparin (LMWH), as well as THSWD alone, on the incidence of Deep vein thrombosis (DVT), D-dimer levels, prothrombin time (PT), activated partial thromboplastin time (APTT), visual analogue scale (VAS) pain score, and calf swelling in patients undergoing hip fracture or replacement surgery, compared to LMWH. Methods: According to the predefined inclusion criteria, we conducted a comprehensive search for randomized controlled trials (RCTs) examining the efficacy of THSWD combined with LMWH or THSWD compared to LMWH in patients with hip fractures or undergoing replacement surgery. The search was performed across multiple databases including China National Knowledge Internet, WanFang, Sinomed, Duxiu, PubMed, Embase, Google Scholar, Cochrane, and Web of Science from their inception until December 2023. Additionally, relevant literature references were retrieved and hand searching of pertinent journals was conducted. The methodological quality assessment of the included trials was carried out following the guidelines outlined in the Cochrane Handbook. Review Manager 5.4 was applied in analyzing and synthesizing. Results: A total of 18 RCTs with 1353 patients were included. The results of meta-analysis showed that compared with the control group, the combined group had a better effect on the incidence of DVT [RR = 0.32, 95% CI(0.17, 0.58; P = .0002], D-dimer [SMD = −5.88, 95% CI(−7.66, −4.11); P < .00001], VAS [MD = −1.16, 95% CI(−1.81, −0.50); P = .0005], Calf circumference difference [MD = −0.56, 95% CI(−1.05, −0.08); P = .02]. There was no significant difference in PT and APTT between the combined group and the control group. Meta-analysis results show that the D-dimer, incidence of DVT, PT, and APTT did not significantly differ between the THSWD and the LMWH groups. Conclusion: This meta-analysis shows that compared with LMWH, THSWD combined with LMWH has a better efficacy in the treatment of DVT after hip surgery, without a significant increase in the incidence of adverse events. Additionally, the combined therapy can also reduce D-dimer, VAS, and swelling. However, due to the limitations of the included studies (such as small sample size and low-quality evidence), the results need to be further verified in more rigorous multicenter clinical trials with a large sample size.

A trombose venosa profunda é uma importante causa de morbimortalidade no mundo, principalmente no âmbito intra-hospitalar, justificando seu tratamento e também profilaxia quando necessário. Os métodos de indução de trombose venosa profunda em animais são fundamentais para o estudo da fisiopatologia da doença, assim como para testes de drogas antitrombóticas. Os objetivos do trabalho foram avaliar e comparar dois métodos de indução de trombose venosa profunda em ratos: o modelo por estase venosa, amplamente descrito na literatura, e o modelo por lesão endotelial, com poucos estudos, principalmente em ratos. Foram usados ratos machos Wistar para indução da trombose venosa profunda. Para estase foi dissecada e ligada a veia cava inferior por 3 horas e após retirado o segmento com a veia contendo o trombo. Para o modelo de lesão endotelial, foi aplicado por 1 minuto um pedaço de papel filtro embebido em FeCl3 e avaliado o segmento após 1 hora. Em ambos os modelos foram avaliados o peso úmido e a área de oclusão. Para o peso úmido obteve-se comparando os métodos de lesão endotelial e estase respectivamente: 17,7mg (±3,0mg) vs. 2.34mg (±1,8mg) com P<0,001. Para a área de oclusão obteve-se comparando os métodos de lesão endotelial e estase respectivamente: 85,11% (±9,67%) vs. 40,83% (±33,14%), com P<0,05. Em todas as variáveis o método de lesão endotelial possuiu resultados superiores ao método de estase venosa, mostrando ser mais reprodutível.
