DEAD-box helicase DP103 defines metastatic potential of human breast cancers

Abstract: Supplemental Table 3A. Importantly, much like DP103, the highest levels of MMP9 were found in primary breast tumors that showed metastasis compared with nonmetastatic or benign tumors (Figure 3E). Importantly, the correlation between DP103 and MMP9 a strong positive clinical correlation between expression of DP103 and MMP9 (P < 0.001 in both cohorts). To further substantiate this finding, we next investigated whether a similar correlation between DP103 and MMP9 expression existed in the cohort from 1-3) or pB… Show more

“…Luciferase reporter assay A luciferase assay was performed as described previously (84). Briefly, 293T HEK (human embryonic kidney) cells were seeded at a density of 3 × 10 4 cells per well in 24-well plates and transfected the following day using Lipofectamine LTX (Life Technologies, Thermo Fisher Scientific) with 0.75 μg pGL3 reporter plasmids and TERT or RPC32-expressing plasmids, along with 30 ng Renilla luciferase constructs.…”

“…Among the various pathways involved in controlling cytokine regulation, the NFκB signaling pathway is widely recognized and studied. NFκB is a principal transcription factor that controls various biological processes, including corneal wound healing, inflammation20, angiogenesis, apoptosis and the stress response2122232425. NFκB is also thought to be involved in several ocular surface disorders, including chemical injury, microbial infections, dry eye, pterygium, ultraviolet radiation-induced injury and corneal graft rejection26.…”

“…NF-κB is a transcription factor well known for regulating many cellular pathways including tumorigenesis 17,18 . Although NF-κB signalling can regulate TERT expression in vitro 9 and two potential NF-κB-binding motifs have been identified within the TERT promoter 19 , regulation of TERT by NF-κB in human cancers is not documented.…”
