BackgroundStevia rebaudiana (Asteraceae), native from Paraguay, accumulates steviol glycosides (SGs) into its leaves. These compounds exhibit acaloric intense sweet taste which answers to consumer demands for reducing daily sugar intake. Despite the developpement of S. rebaudiana cultivation all over the world, the development of new cultivars is very recent, in particular due to a colossal lack of (1) germplasm collection and breeding, (2) studies on genetic diversity and its structuring, (3) genomic tools.ResultsIn this study, we developped 18 EST-SSR from 150,258 EST from The Compositae Genome Project of UC Davis (http://compgenomics.ucdavis.edu/data/). We genotyped 145 S. rebaudiana individuals, issued from thirty-one cultivars and thirty-one landraces of various origins worldwide. Markers polymorphic information content (PIC) ranged between 0.60 and 0.84. An average of 12 alleles per locus and a high observed heterozygoty of 0.69 could be observed. The landraces revealed twice as many private alleles as cultivars. The genotypes could be clustered into 3 genetic populations. The landraces were grouped in the same cluster in which the oldest cultivars “Eirete” and “MoritaIII” type are also found. The other two clusters only include cultivated genotypes. One of them revealed an original genetic variability. SG phenotypes could not discriminate the three genetic clusters but phenotyping showed a wide range of composition in terms of bitter to sweet SGs.ConclusionThis is the first study of genetic diversity in Stevia rebaudiana involving 145 genotypes, including known cultivars as well as landrace populations of different origin. This study pointed out the structuration of S. rebaudiana germplasm and the resource of the landrace populations for genetic improvement, even on the trait of SG’s composition.

Stevia (Stevia rebaudiana Bertoni) is a perennial crop belonging to the Asteraceae family. Its main agronomic and industrial interest is the accumulation in its leaves of steviol glycosides (SG), natural acaloric sweeteners which are 300 times sweeter than sucrose. In order to expand phenotyping knowledge on this emerging crop, researchers and breeders need a universal scale to describe the development of this plant. The objective of this study was to describe the phenological growth stages of stevia through the use of the Biologische Bundesanstalt Bundessortenamt and Chemical Industry (BBCH) scale to provide a standard scale for stevia grown under temperate conditions of production and climate. The observation of phenological stages was performed on genetic resources and validated on stevia progenies grown in field conditions. The 10 phenological stages, namely germination, bud development, foliar development, side shoots appearance, stems elongation, development of harvestable part of the crop, inflorescence emergence, flowering, development of fruit, seeds ripening and senescence, were used to describe the stevia, supplemented with pictures. The chronology and duration of each stage were also observed and described according to the accumulation of degree days during the growing season. This study presents the first BBCH scale established for S. rebaudiana cultivated in temperate conditions and a chronology of phenological stages based on the accumulation of thermal time, through growing degree day calculation (°C/day). This work provides a tool that can be widely used by farmers, breeders and researchers leading to better agronomic management and scientific communication.

Heritability and genetic variance estimates for agronomic traits and glycoside yield in four elite stevia breeding populations

Heritability estimates of traits assist breeders in optimizing cultivar development and can help predict gain from selection in breeding programs. Narrow-sense heritabilities were measured in agronomic traits and glycoside concentration in four elite stevia (Stevia rebaudiana Bertoni) populations using half-sib family analysis. Gain from selection at 20% selection intensity was estimated for morphological, yield, and glycoside traits. Experiments were conducted in two field locations in North Carolina.Variation was observed across cultigens for agronomic traits including stem height, branch width, and leaf size, for both subjective and objective measurements. Yield, lodging resistance, disease resistance, and percent survival showed a high genetic variation across cultigens. In addition, glycosides including Reb A, Reb C, and Reb D also showed high variation by cultigen. Heritability estimates for plant morphological traits were highest for stem height, and moderate for yield and branch width.However, other traits such as leaf area and lodging resistance showed low heritability (0.10). Glycosides including Reb A, Reb C, Reb D, stevioside and total steviol glycosides (TSG) showed high heritability. The estimated gain from selection per year for stem height (24 mm/year), leaf area (131 mm 2 /year) and yield (dry weight) (1.2 Mg/ha per year) suggest significant improvements are possible per breeding cycle. The estimated gain for glycoside concentration and percent of TSG of Reb A (26.44 mg/g, 14.51%), Reb C (20.20 mg/g, 20.17%), Reb D (1.70 mg/g, 1.49%), stevioside (6.73 mg/g, 10.72%), and TSG (20.90 mg/g), similarly, indicate significant gains are possible per breeding cycle.
