A protein associated to prodigiosin formation was found in Serratia marcescens. The protein was not found in nonpigmented strains and was correlated with the pigment level. The protein was about 100 kilodaltons (kDa) and was also found in nonpigmented bacteria of the pigmented strain grown in glucose medium, at high temperature, or under anaerobic condition. The 100 kDa protein was found not in the outer membrane and the periplasm, but in the inner membrane and/ or the cytoplasm. The protein was also found singly or dominantly in pigmentprotein complexes and pigment-localizing vesicles described in previous reports. These results suggest that the 100 kDa protein is associated with prodigiosin formation.A characteristic red pigment, prodigiosin, is found in typical wild-type strains of Serratia marcescens. The pigment is a secondary metabolite (21) and is synthesized in the postexponential phase (18). The genes for pigment synthesis are suggested to be encoded on the chromosomes (3). The formation of the pigment is influenced by medium composition, pH, incubation temperature, and oxygen supply (1,7,19). Oxidative metabolism seems to be essential for pigment synthesis (22), but it was reported that ATP inhibits pigment synthesis (11) and that pigmented bacteria are decreased in respiratory activity (9). The mechanism or regulation of the pigment formation is unclear. Aerobiosis and anaerobiosis induce different proteins detected by electrophoresis in Escherichia coli, such as tricarboxylic acid cycle enzymes and glycolytic pathway enzymes, respectively (16,17). Therefore, we first searched for proteins that might be associated with prodigiosin formation and found a characteristic protein in pigmented bacteria.
MATERIALS AND METHODSStrains and culture conditions.
