Defining Specific Cell States of MPTP-Induced Parkinson’s Disease by Single-Nucleus RNA Sequencing

Abstract: Parkinson’s disease (PD) is a neurodegenerative disease with an impairment of movement execution that is related to age and genetic and environmental factors. 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) is a neurotoxin widely used to induce PD models, but the effect of MPTP on the cells and genes of PD has not been fully elucidated. By single-nucleus RNA sequencing, we uncovered the PD-specific cells and revealed the changes in their cellular states, including astrocytosis and endothelial cells’ absenc… Show more

“…More recently approaches using RNA sequencing (RNA-seq) offer good alternative to examine global gene expression changes in disease state. Several PD studies have employed the RNA-seq technique to understand gene expression changes in the MPTP mouse model (Yang et al 2021;Tong et al 2022;Guo et al 2022), patient brain regions (Keo et al 2020;Aguila et al 2021;Tiklová et al 2021), blood from PD patients (Soreq et al 2015;Infante et al 2016;Moni et al 2019), and iPSCs derived from PD patients (Booth et al 2019;van den Hurk et al 2022).…”

“…FFPE tissues transcriptomic mainly focus on large sample RNA-seq analysis 3 and spatial transcriptome research based on micro-region (LCM, Pick-Seq) [4][5][6] or gene chip with spatial barcode (10x visium) 7 . Single-cell RNA sequencing (scRNA-seq) technology had emerged as the most powerful instrument for assessing cell-type heterogeneity 8 . Recently, a study combined scRNA-seq and spatial transcriptomics to achieve gene expression in FFPE had been reported 2 .…”
