“…A direct E 1 S RIA method, similar to those employed in the DSL-5400 kit, was reported previously [20] but the interference of DHEAS in E 1 S determination is not provided in this paper. Interference of DHEAS in many direct immunoassays for testosterone in male and female plasma has also been reported previously, due to the considerably higher plasma levels of DHEAS than testosterone, associated with insufficient specificity of the antitestosterone antibody employed [21]. A very recent paper [22] comparing LC-MS/MS with the DSL-5400 kit also reported higher E 1 S using the kit and suggested the interference of DHEAS, but do not proved it.…”

“…The extraction steps were discarded to streamline the process but the resultant direct methods, although convenient, could suffer significant interference from other steroids. For example, the most abundant serum steroid DHEAS (mM concentration) will interfere with IAs looking for low level androgens such as testosterone (!10 nM) (18). Before the introduction of large analyser systems, there was scope to change the kit supplier if the assay performed badly, but this is now virtually impossible because reagents are tied into the analyser and kits are not interchangeable between different manufacturer systems.…”

“…Faster direct immunoassays replaced the assays with pre-treatment in clinical routine analysis within the last 25 years. Although these direct immunoassays are simple, rapid and cheap, they may lack in reliability, sensitivity and specificity especially in low concentration ranges [4][5][6]. At present, the use of LC-MS/MS becomes increasingly important in clinical laboratory diagnostics.…”
