Delayed Dissolution and Small Molecule Release from Atomic Layer Deposition Coated Electrospun Nanofibers

Abstract: nanofi ber coating technique shows great potential as a method to tune shelflife, mat degradation, and small molecule release from highly water-soluble polymers, hitherto unexplored, in a wide range of fi elds, including biomedical, agricultural, and packaging.

“…cumulative dissolution, thus suggesting a total dissolution over several days. Such a dramatic retardation of dissolution by Al 2 O 3 coatings has also been shown by Vogel et al 35 cycle-coated one. Since the dissolution rates scale with the number of cycles, or in other words with the coating thickness, it is likely that water accesses the nanofiber through defects or bulk transport through the ALD coating.…”

“…Vogel et al demonstrated that ALD can be utilized to slow the mat degradation in both humid and aqueous conditions, from several minutes to multiple weeks, just by controlling the thickness of the deposited Al 2 O 3 coating on electrospun poly (vinyl alcohol) nanofibers through increased ALD cycles [ 212 ]. Thus, changing the rate at which nanofibers dissolve modulates the release of embedded small molecules (ketoprofen) within the polymer matrix from minutes to weeks while reducing the “burst” effect [ 212 ].…”

“…Due to the diffusive and self‐limiting nature of its growth mechanism, this technique allows for sub‐atomic control of layer thickness, in this being superior to the other vapor‐phase methods. Vogel et al reported the treatment of poly(vinyl alcohol) (PVA) electrospun nanofibers loaded with a small molecule drug, ketoprofen, with ALD Al 2 O 3 . The morphology and drug release were monitored as a function of the ALD Al 2 O 3 thickness, ranging from 2 cycles to 200 cycles (Figure ).…”

“…Next to polymer encapsulation, very recently vapor‐phase deposited metal‐oxides have been adopted as drug release barriers, either directly deposited on the drug crystals or on drug loaded polymers …”
