Deletion of the Anaerobic Regulator HlyX Causes Reduced Colonization and Persistence ofActinobacillus pleuropneumoniaein the Porcine Respiratory Tract

Abstract: Actinobacillus pleuropneumoniae, the etiological agent of porcine pleuropneumonia, is able to persist on respiratory epithelia, in tonsils, and in the anaerobic environment of encapsulated lung sequesters. We have demonstrated previously that putative HlyX-regulated genes, coding for dimethyl sulfoxide (DMSO) reductase and aspartate ammonia lyase, are upregulated during infection and that deletions in these genes result in attenuation of the organism. The study presented here investigates the role of HlyX, the… Show more

“…Since other pathogens require Anr homologs for virulence (24)(25)(26)(27), such an approach may also be beneficial in the treatment of other diseases. Future work will determine if the inactivation of Anr during established infections can limit disease.…”

“…Anr homologs have been linked to pathogenesis in other species, including Shigella flexneri (24), Bordetella pertussis (25), and Actinobacillus pleuropneumoniae (26). In Neisseria meningitidis, deletion of two operons directly controlled by the Anr homolog Fnr resulted in attenuated virulence of the pathogen in an infant mouse model of infection (27).…”

“…While required for anoxic growth via denitrification, P. aeruginosa ⌬anr strains are not impaired in growth under hypoxic (low-oxygen) conditions (18,19). Anr homologs have been identified as regulators of virulence in other Gram-negative microbes (20)(21)(22). The high-level expression of transcripts encoding the denitrification and arginine fermentation machinery, as well as certain high-affinity cytochrome oxidases (23), suggests that Anr activity is high in vivo (11,18,24).…”

“…further attenuate this strain, we initially focused on enzymes involved in anaerobic respiration which have been shown to facilitate the pathogen's persistence in the reducing environment of the epithelial lining fluid as well as in necrotic lung tissue with reduced oxygen tension (4). To avoid problems with antibiotic resistance and undefined mutations present in experimental live attenuated A. pleuropneumoniae vaccines described previously (17,27,40), the single-step transconjugation system was employed, as it allows the construction of strains with multiple isogenic mutations not containing foreign DNA as shown in this study.…”
