Delivery of ziconotide to cerebrospinal fluid via intranasal pathway for the treatment of chronic pain

Abstract: The purpose of the current study was to investigate the plausibility of delivery of ziconotide to the cerebrospinal fluid (CSF) via intranasal administration. Ziconotide was administered either in the form of solution or Kolliphor P 407 gels (KP 407) intranasally in Sprague-Dawley rats. The effect of incorporation of chitosan in the formulation was also investigated. Time course of drug in the CSF was investigated by collecting CSF from cisterna magna. Pharmacokinetics of ziconotide in CSF following intratheca… Show more

“…Chemical penetration enhancers are widely used to improve the topical and transdermal delivery of drugs due to their patient compliance and lower cost [7,12]. In this project, ten penetration enhancers were investigated to improve the permeation of propofol across the porcine full thickness skin.…”

“…3D printing is used to produce products like highly porous products (orally disintegrating), toroidal products (approximately zero-order release), Complex drug-device combination products, personalized dosing (for potent drugs and children), drug-loaded implants (matching the anatomy of individual patients), polypills (combination of different drugs) and ocular implant. 3D printing facilitates personalization by changing the digital designs which is more feasible than changing equipment design with less operational cost [10][11][12]. 3D printing is synonymously used with additive manufacturing and rapid prototyping technology.…”

“…Manda et al investigated the ability of delivering ziconotide via intranasal route. Ziconotide when delivered using poloxomer as a vehicle could not only attain therapeutic concentration rapidly but also prolong the release of ziconotide, maintain therapeutic levels in CSF for a longer period of time compared to solution and reduce the frequency of administration [53].…”
