Derivative Recording in Column Chromatography

Abstract: SummaryThe use of an ultraviolet "peak-spotter" and pH-meter to obtain the first derivative of the effluent transmission curve from columns of Sephadex is described. The" method is of use in determining accurate peak separations in effluents, in handling cloudy solutions, as a guide to the fractionation of complex inixtures, and in the study of rapid reversible association in protein systems.
