Description of Aphelenchoides giblindavisi n. sp. (Nematoda: Aphelenchoididae), and Proposal for a New Combination

Abstract: One new and one known species of the genus Aphelenchoides from Iran are studied. Aphelenchoides giblindavisi n. sp. is mainly characterized by having five lines in the lateral fields at mid-body, and a single mucro with several tiny nodular protuberances, giving a warty appearance to it, as revealed by detailed scanning electron microscopic (SEM) studies. The new species is further characterized by having a body length of 546 to 795 μ m in females and 523 to 679 μ m in males, rounded lip region separated from … Show more

“…For reconstruction of SSU and LSU rDNA phylogenies, the homologous sequences of ektaphelenchid and seinurid species were retrieved from the database. The outgroup taxa were selected according to previous studies ( Aliramaji et al, 2018, 2019 ). The selected sequences of both data sets were aligned using Clustal X2 ( http://www.clustal.org/ ) with the default parameters.…”

“…The latter form, however, should not be misinterpreted as a warty mucro, a differentiation at the tail tip of some recently described species of Aphelenchoides , e.g. A. giblindavisi ( Aliramaji et al, 2018 ) and A. hamospiculatus ( Mortazavi and Pedram, 2020 ).As already stated, the tentative/possible synonymies for some species of the genus were recently addressed by Pedram et al (2018) . During the present study, a detailed examination of the original descriptions of L. patulus and L. australis suggested that they could belong to the same species, as the used traits to differentiate the latter species from the former look insufficient (the status of the lateral line in the former species needs further studies).…”

“…The thermal cycling program was as follows: denaturation at 95°C for 4 min, followed by 35 cycles of denaturation at 94°C for 30 s, annealing at 52°C for 40 s, and extension at 72°C for 80 s. A final extension was performed at 72°C for 10 min. Amplification success was evaluated by electrophoresis on 1% agarose gel ( Aliramaji et al, 2018, 2020 ). The PCR products were purified using the QIAquick PCR purification kit (Qiagen®) following the manufacturer’s protocol and sequenced directly using the PCR primers with an ABI 3730XL sequencer (Bioneer Corporation, South Korea).…”
