Design and theoretical evaluation of a novel microfluidic device to be used for PCR

Abstract: The design of a novel, microfluidic chip with an integrated micro peristaltic pump and chambers for DNA amplification is described. This chip contains three reaction chambers stable at 90 • C, 72 • C and 55 • C for PCR amplification, a bi-directional peristaltic pump and optical integrated detection of the droplet. A reactant droplet is to be introduced into the device, pumped back and forth between the chambers by the micro peristaltic pump for sample processing. The static behaviour of the micro pump was mod… Show more

“…The fluid film thickness, given by h + δ i in the presence of diaphragm deflection, does not vary with in-plane coordinates x and y. As shown in the appendix, this greatly simplifies the solution of equation (4). In particular, if the fluid chamber is symmetric about the y-axes, we obtain the hydrodynamic force…”

“…Analytical and numerical calculations of static diaphragm deflections and pumping stroke volumes have been considered for piezoelectric peristaltic micropumps [25]. Also restricted to static characteristics, piezoelectric [4] and electrostatic [26] peristaltic pumps have been modeled by assuming the flow rate to be proportional to the actuation frequency and stroke volume, which is determined from the diaphragm motion by ignoring its interaction with fluid flow. More recently, a lumped-parameter analysis has been reported for diaphragmbased micropumps working peristaltically using active valves driven by general actuation schemes [27].…”

“…(3) (with eqns. (4) and (6) substituting for F e and F h ), (7) and (8). With the chamber model above, the pump can be represented as a system consisting of the three chambers and the inlet and outlet channels leading to and from the pump, as illustrated in Fig.…”

“…In addition, we present a lumpedparameter model to analyze the operation of the peristaltic pump. With the model, the dynamics of peristaltic pumping, not addressed by existing micro pump models, [8][9][10] are appropriately investigated. We consider the dynamics of individual pumping chambers and account for the interactions of such component dynamics by a system representation of the peristaltic pump.…”

“…Piezoelectric materials [1][2][3] which can achieve the conversion between mechanical and electrical energy are widely used in piezoelectric functional devices including sensors, actuators and energy harvesters. Because of the excellent characteristics of the piezoelectric device, it is often used in the field of high technology.…”
