Design of a 13C Magnetic Resonance Probe Using a Deuterated Methoxy Group as a Long‐Lived Hyperpolarization Unit

“…Cells ( 1×10 7 ) were collected in a 15 mL polystyrene tube by centrifugation at 800×g and 4°C for 10 min, suspended in 10 mL of phosphate buffered saline (PBS) buffer, and supersonicated for 1 min. 36) After centrifugation at 800×g for 5 min and 4°C, the insoluble materials were removed, and protease inhibitors were added to the supernatant, which was then frozen and stored at −20°C.…”

“…To address this crucial issue, attention has focused on the development of molecular units that can retain the hyperpolarized state for longer periods101112131415161718. The hyperpolarization lifetime is directly correlated with the spin–lattice relaxation time ( T 1 ) of nuclei.…”

“…In practice, hyperpolarized [ 13 C]bicarbonate9, [ 13 C]benzoylformic acid10, [ 13 C, D 6 ] p -anisidine11 and [ 13 C]dehydroascorbate1213 have been designed as sensitive MR probes for sensing pH, H 2 O 2 , HOCl and redox status, respectively. A universal strategy—in other words, the presence of a platform structure for designing hyperpolarized MR probes—can make it easier to develop a variety of hyperpolarized MR sensors.…”
