Detailed map of a region commonly amplified at 11q13→q14 in human breast carcinoma

Abstract: Amplification of loci present on band q13 of human chromosome 11 is a feature of a subset of estrogen receptor positive breast carcinomas prone to metastasis. As many as five distinct amplification units have been described on 11q13. They include particularly a genomic area encompassing the GARP gene at 11q13.5→q14.1. We have reassessed our current knowledge of this region, located telomeric to CCND1 and EMS1, which is amplified in 7–10% of mammary tumors. The loose definition of the driving forces of these am… Show more

“…One of these, a novel transcript on chromosome 11q13, was the transcript found most frequently. The region 11q13 has long been known to be relevant for tumorigenesis and metastasis (Rasio et al, 1995;Bekri et al, 1997;Chakrabarti et al, 1998). To determine sequences upstream and downstream of the identified clones, we used 5 0 and 3 0 RACE techniques and obtained an RNA sequence of 940 nt.…”

“…Other tumor suppressors play a more direct role in regulating autophagy such as DAPk, 89 Beclin 1, p19ARF (smARF) and UVRAG. [90][91][92][93] p53 falls into this category as well through upregulation of DRAM, which has also been shown to be downregulated in certain carcinomas. 62 Even these proteins, however, may exert their tumor suppressive activity through additional functions.…”

“…146 Mutation of UVRAG is frequently observed in various human cancers and may be responsible for the etiology of these cancers. [147][148][149][150] UVRAG is critical for DNA repair and chromosomal stability, especially in response to radiation. [151][152][153] Using GST affinity isolation, UVRAG was identified as a binding partner of BECN1-PIK3C3, which directly interacts with BECN1 via its CCD.…”
