Detecting Clonal Rearrangement in Non-Hodgkin's Lymphomas in Taiwan by Polymerase Chain Reaction

Abstract: The detection of monoclonal expansions of the immunoglobulin heavy chain (IgH) or the T-cell receptor-gamma (TCRgamma) chain genes is an important supplement for the diagnosis of the non-Hodgkin's lymphomas (NHLs). Detection of monoclonality by polymerase chain reaction (PCR) method has offered an efficient approach for rapid diagnosis and monitoring of the therapeutic effects. Here we conducted a retrospective PCR clonality study on 49 cases of NHLs including 23 B-cell lymphomas (BCLs), 20 peripheral T-cell l… Show more

“…22 For FFPE samples, the sensitivity has been reported to range from 80 to 98% for TCR. [23][24][25][26] ENKTLs are frequently accompanied by necrosis and apoptosis of the tumor cells, leading to degradation of DNA, ineffective PCR reactions, and false-negative results. 27 In a study by Pongpruttipan et al, 8 clonal TCR gene rearrangements were documented in only three out of six TCR protein-positive cases.…”

“…Genomic DNA was extracted from formalin-fixed, paraffin-embedded tissues by using a Qiagen DNeasy Blood & Tissue Kit (Qiagen; Hilton, Germany) according to the manufacturer's instructions. The primers for TCRγ chain gene and the protocol for PCR were designed as previously described [15]. A case of PTCL-NOS was used as positive control, and a normal lymph node as a negative control.…”

“…Although previously reported frequencies of B-cell clonality in AILT are similar, 20,23,32,33 those for PTCL range from 9 to 16%. 2,4,5,22,34,35 We considered that some cases of PTCL-NOS were misclassified and actually represent AILT. However, PTCL-NOS cases did not have classic morphologic features of AILT.…”
