Detecting the Enterotoxigenicity of Staphylococcus aureus Strains

Abstract: An optimal sensitivity plate method for examining large number of staphylococcal strains for production of the known enterotoxins (A—E) is presented. Small volumes of relatively concentrated enterotoxin are produced by the semi-solid agar, cellophane-over-agar, or sac culture techniques. Detection of the enterotoxin in the supernatant fluid is accomplished with the optimal sensitivity plate method. In this method small plastic petri dishes (50 mm) were used for a modified Ouchterlony of high sensitivity.

“…To visualize the ability of producing SEA of the 109 selected cultures, OSP (Optimum Sensitivity Plate) immunodifusion method was used (ROBBINS et al, 1974). The immunodifusion medium was prepared with 8.5g of NaCl, 5.0 mL of mertiolate solution at 2% and 12.0 g of high purity agar dissolved in 1000 mL of Tris buffer 0.05M, ph 7.4.…”

“…Immunodiffusion assay. Gel double-immunodiffusion assays of rTSST-1 and mTSST-1 with anti-TSST-1 antibody were done basically according to the method described by Robbins et al [41], with minor modifications. Noble agar (Difco), 1.2% in 0.01 mol/L PBS, which contained 10 ppm thimerosal, was poured into plastic petri dishes.…”

“…Serological assays. Western blot (immunoblot) analysis and gel double-diffusion assays were performed as described previously (27,50). Gel double-diffusion-grade SEA and rabbit polyclonal anti-SEA and anti-SEB antisera were obtained from Merlin Bergdoll.…”
