Detection, Identification and Molecular Characterization of a Phytoplasma Associated with Arabian Jasmine (Jasminum sambac L.) Witches’ Broom in Oman

Abstract: Arabian jasmine (Jasminum sambac L.) plants showing witchesÕ broom (WB) symptoms were found in two regions in the Sultanate of Oman. Polymerase chain reaction (PCR) amplification of the 16S rRNA gene and the 16S-23S spacer region utilizing phytoplasmaspecific universal and designed primer pairs, and transmission electron microscopy of phytoplasma-like structures in phloem elements confirmed phytoplasma infection in the symptomatic plants. PCR products primed with the P1/P7 primer pair were 1804 bp for jasmine … Show more

“…In China and Vietnam it is used to produce the flowers used in jasmine tea and is grown in plantations. There are relatively few reports of disease of jasmine; these include witches' broom caused by phytoplasma (Al-Zadjali et al 2007), tomato mosaic virus infection (Kamenova et al 2006), cercosporoid leaf spots (Braun and Sivapalan 1999) and several other folicolous taxa (Agarwal and Sahni 1965;Sahni 1966). Reports of Colletotrichum species on jasmine are rare.…”

“…Phytoplasma asteris' in Lithuania (Jomantiene et al 2000) caused alfalfa witches'-broom disease. In Asia, phytoplasmas of the 16SrII-D subgroup in Oman (Khan et al 2002, Alzadjali et al 2007, the elm yellows group in China (Li et al 2012) and a mycoplasma-like organism in Saudi Arabia (Cook & Wilton 1985) were identified as causal agents of alfalfa witches'-broom. Moreover, 'Ca.…”

“…Plants infected with phytoplasmas exhibit a range of symptoms such as virescence, phyllody, yellows, witches’ broom, leaf rolling and general decline (Bertaccini et al. 2005) and also stunting, floral abnormalities, shoot proliferation, yellowing of foliage and reduced vigour, often leading to the death of the plant (Al‐Zadjali et al. 2007).…”
