“…Dashed horizontal line : limit of enumeration in water ammonium acetate precipitation followed by further purification using SpinBind columns was found to be highly effective for removal of humic contaminants. Detection limits of less than 1 cfu ml-' water have been reported using non-selective media (Maheshkumar et al 1990;Ford 1994) but for the samples used in this study the limits were more in agreement with those found by Morgan et al (1993) of around 300 cfu ml-'. Even with the aid of catechol spraying, accurate enumeration on permissive media was not possible at cell densities of < lo4 ml-' (day 3 onwards) due to overgrowth by the indigenous microflora.…”

“…This research was originally motivated by a need for nonlethal sampling procedures to evaluate disease status in valuable threatened or endangered fish. Procedures developed by Ford (1994) to isolate A. salmonicida from the water and those de- scribed by Cipriano et al (1992) to isolate the pathogen from mucus enable hatchery biologists to detect A. salmonicida at early states of infection. When incorporated into a regular fish health monitoring program, these procedures provide biologists with proactive tools to minimize the deleterious effects of disease.…”

“…Bacteria were quantified in dilutions containing 10-30 colony-forming units (cfu). In addition, water samples from the White River (the source of water for the culture units) were processed weekly for the presence of the bacterium as described by Ford (1994). Individual bacterial colonies derived from water and fish samples were subcultured onto tryptic soy agar (Difco Laboratories, Detroit.…”

“…Les analyses bactériologiques ont été faites suivant la technique des membranes filtrantes (HAVELAAR ef al., 1987 ;FORD, 1994), en utilisant des membranes en ester de cellulose -Millipore, Bedford, MA 01730 -de porosité 0,45 nm. La recherche et l'isolement de P. aeruginosa ont été faits sur milieu Pyocyanosel (MARCHAL et al, 1991).…”
