Detection of fractalkine in human seminal plasma and its role in infertile patients

Abstract: These results suggest that fractalkine is a chemokine associated with sperm motility and the migration of CX(3)CR-positive leukocytes into semen.

“…CXCL10 is able to inhibit progesterone synthesis in Leydig cells and proliferation of these cells 54 . The effect of CXCL8 on spermatozoal motility is unclear, but the high seminal plasma CXCL8 concentration was reported in the men with leukocytospermia 56 as well as the low CX 3 CL1 concentration in the subfertile men with asthenozoospermia 57 . These findings are in consistent with the results in murine autoimmune orchitis models 58 .…”

“…In recent years, several reports have attempted to correlate different SP protein levels and semen parameters, such as sperm concentration, motility, and morphology (Baumgart et al 2002, Zhang et al 2002. We followed this objective in order to determine the possible diagnostic validity of MSMB concentration as a biochemical marker that could provide useful information on male fertility potential.…”

“…RT-PCR was performed using an RT-PCR high kit (TOYOBO Co., Osaka, Japan), as previously described [10][11][12]. Amplification using CX 3 CR1-specific primers yielded a 653-bp DNA product, while amplification using the fractalkine-specific primers yielded a 597-bp DNA product (5).…”

“…CX 3 CR1-mediated signal transduction presumably plays a role in cell migration and adhesion [8,9]. In the reproductive system, this molecule is present in the seminal plasma and is associated with sperm motility [10]. We reported that fractalkine contributes to the immunodefense mechanism during pregnancy [11] and that this molecule is present in the Fallopian tube and that its receptor exists in the spermatozoa [12].…”
