Detection of gp43 of Paracoccidioides brasiliensis by the loop-mediated isothermal amplification (LAMP) method

Abstract: Paracoccidioidomycosis is a deep mycosis caused by the thermo-dependent dimorphic fungus Paracoccidioides brasiliensis and is prevalent in Latin American countries. We detected the species specific gp43 gene of P. brasiliensis by loop-mediated isothermal amplification (LAMP) in 22 clinical and seven armadillo-derived isolates. The amplified DNA appeared as a ladder with a specific banding pattern. The advantage of the LAMP method is speed; only 3 h were necessary for identification of the organism and diagnosi… Show more

“…3A). This sensitivity is comparable to that of standard PCR and similar to values (10 to 100 fg) mentioned by other authors for their LAMP-based systems (7,17,34). However, the LAMP detection limit was only 1 pg when C. krusei PYCC 3341 T genomic DNA was used (Fig.…”

“…For instance, we obtained better results in the LAMP reaction with a 90-min incubation at 64°C than with incubation for a standard period of 60 min or less. Some authors would corroborate this result (7,8,36), while others would disagree (29). Another possibility for increasing LAMP sensitivity and accelerating the response time would be the additional utilization of loop primers in the reaction mixture (31).…”

“…An accurate and rapid diagnosis system for periodontitis is essential for periodontal treatment. Several recent reports have demonstrated the usefulness of the LAMP method (10,11,17,26,27,33,49). Therefore, we focused on the LAMP method for the rapid detection of periodontopathic bacteria.…”

“…Each aliquot of 200 µL of swab specimens was tested by the LAMP method, as described above. On the other hand, aliquots of 20 methods are available Endo et al, 2004;Kawano et al, 2015;Ohori et al, 2006;Uemura et al, 2008;Yo et al, 2016 . In this study, we developed a LAMP method using a primer set for detecting a wide range of fungi from environmental specimens to evaluate the fungal burden, which should help to prevent allergies or infections caused by fungi.…”
