Detection of histamine in beer by nano extractive electrospray ionization mass spectrometry

Abstract: In this study, rapid quantitative detection of histamine in beer was achieved by using nano extractive electrospray ionization mass spectrometry (nano EESI-MS) coupling with standard addition method. Based on the MS(2) experiment, histamine concentrations in three beer samples were determined to be 1.10 ± 0.12 µg/ml, 0.81 ± 0.09 µg/ml and 0.79 ± 0.09 µg/ml. The limit of detection for this method was calculated to be 0.02 µg/ml. These results show that this novel method can be used for direct, rapid and sensiti… Show more

“…The content of the target dipeptides in hydrolysate must therefore have been over-estimated when the absolute standard calibration curve was used. To avoid interference from matrix effects in the MS analysis of food products, a study similar to ours was performed by Cai, Li, Xiong, Fang, and Xu (2013). They demonstrated the accurate quantitative analysis of histamine in beer by the standard addition method, coupled with an extractive nano-ESI-MS technique, without the requirement for matrix cleaning.…”

“…Ambient ionization techniques, such as SESI [24][25][26] and EESI [27][28][29] have demonstrated high sensitivity for gas phase and condensed phase analytes. The coupling of GC with ESI can be considered as the coupling of a separation technique with SESI/EESI leading to improved analytical performance.…”

“…Mass spectrometry (MS) is increasingly applied in the analysis of histamine and related biogenic amines because of its significant advantages: sensitivity, selectivity and rapid analysis. [9][10][11][12] Chen and coworkers have used neutral desorption extractive electrospray ionization mass spectrometry for the rapid in-vivo analysis of histamine and other biologic amines in frozen meat without sample preparation and separation. 10,11 However, such direct MS detection of small molecules in complex matrices often suffers from large background noise in the low mass range (< 150 Da) which is caused by the species and their fragments of the matrices.…”
