Detection of human cytomegalovirus genome and gene products in central nervous system tumours

Abstract: Human cytomegalovirus (HCMV) genome and related proteins have been reported in a great proportion of malignant gliomas. However, these results are unexpected since HCMV is not known as an oncogenic virus. By immunohistochemistry (with an anti-IE1 monoclonal antibody) and in situ hybridisation (with biotinylated DNA probes) on tissue microarrays and frozen sections, we investigated a French series of central nervous system (CNS) tumours, including 97 glioblastomas. In 10 cases of glioblastoma, rare astrocyte-li… Show more

“…Indeed, one group only has reported on the detection of this virus in CNS tumours and also in colonic and prostate cancers (Samanta et al, 2003). As far as the CNS tumours are concerned, we have been unable to reproduce the results of the study by Cobbs et al We have found that the virus could be detected in only a few cases of CNS tumours not in malignant cells but rather in putative stromal cells (Sabatier et al, 2005; data not shown).…”

“…Furthermore, formalin-fixed, paraffin-embedded samples were obtained from two adult patients with HCMV reactivation. Cell lysates from fresh frozen tissues from patients #1, 3,4,8,18 in Table 1 were applied for immunoblotting. Paraffin sections from patients #41-50 and #P3 in Table 1 were applied for immunohistochemistry and FISH.…”

“…In contrast, Sabatier et al (2005) screened 132 CNS tumours, including 97 GBMs, by IMH (anti-IE1 monoclonal antibody) and ISH with tissue microarrays and frozen sections and found rare astrocyte-like cells staining positive for HCMV in only 10 cases of GBM and questionable staining of rare cells in one case. This result suggested that none of the CNS tumours contained HCMV genomes and/or proteins in a significant proportion of tumour cells.…”
