Determinants of leptin gene expression in fat depots of lean mice

Abstract: The relationship of leptin gene expression to adipocyte volume was investigated in lean 10-wk-old male C57BL/6J mice. mRNA levels for leptin, insulin receptor, glucocorticoid receptor, and tumor necrosis factor (TNF)-α in inguinal, epididymal, and retroperitoneal adipose tissues were quantified and related to adipocyte volume. Leptin mRNA levels were highly correlated with adipocyte volume within each fat depot. Multiple regression analysis of pooled data from the three depots showed that leptin mRNA levels we… Show more

“…It is well known that leptin levels strongly correlate with the amount of lipid 37,38 and its release is proportional to adipocyte volume. 36 Compared to the uninfected control, the virus increases differentiation of 3T3-L1 cells, 11 resulting in greater proportion of adipocytes and the lipid content. As adipocytes, but not preadipocytes secrete leptin, it would be misleading to compare leptin secretion of two groups containing unequal number or size of adipocytes.…”

“…36 Considering the continued relative suppression of leptin expression by Ad-36 on day 5 (Figure 2), and to allow its adequate production and release, leptin release by 3T3-L1 cells was determined on day 5 post-inoculation (Experiment 2). Leptin levels were within the detectable range of the standard curve.…”

“…It has been postulated as a sensor of fat mass and a hormonal signal that exerts its effect on energy regulation through its hypothalamic receptor (15,16). Because the adipocyte is the only source of ob gene products, serum leptin level has a strong positive association with the amount of body fat and adipocyte leptin mRNA in humans as in rodents (35)(36)(37). Several studies have examined serum leptin level and future weight gain, but few have examined change in regional adiposity (17,38,39).…”

“…It has also been discussed whether the regional difference in leptin gene expression might be due partly to a difference in mean fat cell volume (8). In lean mice, depot-specific differences in leptin gene expression are mainly related to the volumes of the constituent adipocytes (25).…”
